Ataxia telangiectasia-mutated protein kinase (ATM) : 毛细血管扩张性共济失调突变蛋白激酶

是一种PI3K样的丝氨酸/苏氨酸激酶,包含有PI3K结构域和Rad3结构域,参与了激活细胞周期关卡,控制端粒长度和应答DNA损伤等生物学过程。 DNA双链断裂会导致ATM蛋白激酶的激活。可以被ATM磷酸化的下游靶点包括p53和NBS1蛋白以及组蛋白变体H2AX。通过这种方式, ATM可以影响细胞周期的过程和DSB修复以及细胞凋亡。

taxia telangiectasia mutated (ATM), a serine/threonine protein kinase , is a member of PI3K family proteins. ATM is activated by DNA damage. ATM can phosphorylate many kinase such as p53 and CHK2, resulting in apoptosis and cell cycle arrrest. ATM can be retrieved from nuclear extracts by DNA oligonucleotides or by DNA finity chromatography becasue ATM can bind to DNA. The checkpoint clamp proteins may play the role in modulating the signal (via its processing) or the transmission of the signal. The active PIKK then activates and/or recruits the checkpoint kinases of the Chk1/Chk2 families, which act as effector kinases of the DNA damage pathways. DNA damage can be  detected (either directly or after processing) by ATM. In vertebrates, different DNA damage signals elicit either a branch composed of the ATM-Chk2 axis.
